In order to prepare conditioned media from primary cultured dental pulp cells, passage 5 is widely reported. May I have some scientific explanation despite of the inadequate quantity of the cell. Thank you
Human dental pulp stromal cells possess angiogenic potential as they secrete paracrine factors that may alter the host microenvironment.
So, the conditioned media derived from the primary cultured dental pulp cells is basically used to investigate its effect on endothelial cell behavior in vitro. The amount of proangiogenic and anti-angiogenic proteins secreted by these cells is high from passage 4 and declines gradually up to passage 8. So, you would need conditioned media derived from primary cultured dental pulp cells from passage 4 to 6 for your experiments.
