I'm doing isolation and culture of human monocytes from healthy subjects. I'm obtaining PBMCs from whole blood and purifying monocytes using density gradient centrifugation method. After isolation, I'm seeding 1.2 million monocytes per well with 1 ml IMDM (contains penicillin/streptomycin, not containing autologous serum and M-CSF) in a 6 well plate and incubating for 2h. I'm trying to differentiate monocytes to macrophages, so after incubation I'm replacing media with 1 ml IMDM containing autologous serum and 50 ng/ml M-CSF. Cells are culturing for 7 days, media changes occuring at day 1 and day 4.

My problem is my cells are not sticking or cells are detaching, dying day by day and by the end of the 7th day I am not getting enough macrophages. What could be the problem?
