Why I got bulgy structure on the surface of my chitosan scaffold after keeping it in 4 degree for over night and then transfering to deep freezer for rest 2-3 days. I was about to go for lyophillization followed by characterization.
How this will hamper my scaffold's SEM characterization?
Water expands as it freezes. The water freezes from the outside of the tubes to the inside, the water in the middle can only expand by moving up creating the "bulge"

This is also why pipes burst in houses when they freeze- there is no place for the water to expand into, so it splits the pipes when it expands.

If the material was in solution or suspended solid, it should be ok. If it were cross-linked like an aerogel, the material might have been stressed.
