Hi, I am synthesising chitosan nanoparticles using the ionic gelation method and I am characterising the nanoparticles in distilled water and in DMEM (cell culture), however the zeta potential in water is above 17mV where as in DMEM (cell culture media) it is below 5mV. Now I am trying to stabilise these nanoparticles, please advice?
To stabilize the chitosan nanoparticles and maintain their zeta potential in the cell culture medium (DMEM), you can try the following approaches:
1- Optimize the ionic strength of the DMEM to match the distilled water conditions.
2- Adjust the pH of the DMEM to the optimal range for chitosan nanoparticle stability (around pH 5-6).
3- Use stabilizing agents like polyvinyl alcohol (PVA) or polyethylene glycol (PEG) to provide steric stabilization and prevent aggregation.
4-Modify the chitosan properties, such as the degree of deacetylation or molecular weight, or use chitosan derivatives like quaternized chitosan; and 5- Characterize the nanoparticles in the actual cell culture medium to understand their behavior in the relevant biological environment and guide your optimization efforts.
