You can use a trypsin solution to detach the cells on the bottom of the flask and proceed normally.

My question is, this cell line (M-NFS-60) are not in adherent nature. However, this cells are also not give response in assay. I am suspecting its because of changes in its inherent property.

Maybe your G-CSF lever used in cell culture is too low, I have used this cell line for about one year, and cells remain grow in suspension. The concentration of h_M-CSF I used in my cell culture is 60ng/ml. By the way, why did you change M-CSF with G-CSF?

I know I should adapt this cells with MCSF but we adapted this cells deliberately with GCSF considering that the cells are going to use for GCSF testing only.
