I have a reporter that expresses GFP from an E2F1 promoter in my T98G cells (glioblastoma). Between technical replicates, I'm able to reproduce the same results nicely, however between biological replicates (difference in time about 1 week) I can get pretty different results. I'm curious if there's something I'm missing because I'm conducting the same experiment each week and getting different results every time.
You can have a few issue. It is natural in the case of biological replicas. I suggest use an internal standard, whom response would be the reference in the given experiment (I suppose you referenced absolute numbers considering in diff). Some of the problems probably came from TC issues. Try a Hepes-buffered medium if it is not your case..
