What is the rationale behind the administration of intracerebroventricular streptozotocin (ICV-STZ) in a dose of 3 mg/kg twice (day 1 and day 3) in rats to produce an animal model of Alzheimer's disease? Can the same level of memory impairment will be generated if the dose was used only once (day 1) instead of twice. The mortality rate will be affected or not? Further, I would like to ask, should the animals be recovered completely from stereotaxic surgery to start treatment (oral or ip)?

STZ infused intracerebroventricularly to rodents in a subdiabetogenic dose and induce the AD in a dose dependent manner. In our lab we performed a pilot study with a single dose of ICV STZ (to avoid alternate dose and opening of suture) and found a memory impairment but no Abeta deposition in hippocampus. Then we followed a protocol of available literature and infused animals with two alternate doses. Then a memory impairment with appearance of Abeta in hippocampus was found. The mortality rate in ICV STZ model is 15-20%. Secondly, the animal gets recovered after proper care and use of topical antibiotics. But the drug treatment should be started after 1 or 2 days of ICV STZ treatment.
