Why doesn’t the FRAP reagent + sample turn blue?

I have prepared the FRAP reagent according to Benzie & Strain (1996). My samples are essential oil nanoemulsion. However, after adding the FRAP reagent into the sample, it doesn’t turn blue. How do I rectify this? I have alrady tried preparing fresh FRAP reagent and fresh samples several times yet no difference. But when used with FeSO4 for standard curve, it turned blue right after adding FRAP.

I think you should try to prepare your samples in an emulsifying agent (Tween or DMSO) because FRAP is a hydrophilic solution. In contrast, essential oil is lipophilic, so the reaction will not occur. I also recommend preparing several concentrations of your sample in order to determine the appropriate concentration for your calculations.
