some literature stated that colonization rate of endophytic fungi varies in the thickness of the plant but the literature did not stated why colonization rate varies. does nutrient distribution, absorption of endophytic fungi is one of the reasons? the leaf I used in my study was Cordia dichotoma Forst G. (which has a thin leaf), regarding its endophytic fungi and its activity of antimicrobial, but there are no Researches yet (base on the internet; e.g. elsivier, science direct, springer about it and its endophytic fungi some Review articles about the plant too didn't stated that there are studies about it.
First of all, if you want to isolate endophytic fungi , in Culture dependent method, the most important Parameter is leaf Surface disinfection, that is related to leaves thickness, because in Thin leaf , if you use normal disinfection may be you kill endophyte too, and if not maybe some ephyphytes grow, i suggest to increase your repitation with normal disinfection if your aim is isolation, generally diversity and Population of endophyte in leaf is not related to thickness.
Hi: I think because the endophytic fungi not produce a toxic secondary metabolite that kill the tissue of the plant like pathogenic fungi, so these endophytic fungi can not colonization the deep tissues or dont have the mechanism to go deep in thickness tisse (like lysis enzyme).
