My sample is polystyrene microspheres dispersed in Phosphate-buffered saline( PBS ). Then I use an injection pump to force the sample flow through a microchannel(shows in the figure as follows). The polystyrene microspheres are not visible in the PBS which flows through the channel. Why?
Because of sedimentation? the density of polystyrene microspheres is about 1.05g/cm3. The diameter of the polystyrene microspheres is 12um. Or something else? thanks for your kind answers.
How does the mixture of PS spheres and PBS look at the same magnification if you just put a droplet on a microscope slide? (I wonder whether the spheres are visible at all in PBS resp. if one had to use light of a certain color or at a certain angle of incidence.)
Joerg Fricke Thanks. It is visible when I just put a droplet on a microscope slide. It seems that increasing the concentration of PS can solve the problem. Of course, high concentration is easy to cause a block in the microchannel.
U. Dreher Thanks for your answer. I don't know the accurate density of PBS(1x) Actually, I added 0.1% tween20 to the solution.
