Why does the concentration of collagen (hydroxyproline) not consistent each extraction?

We have been extracting hydroxyproline from marine animals and the extraction was repeated several times. However, the results were not consistent and sometimes it turned out 0. We had tried the acid extraction as well as pepsin soluble extraction method. Anyone have experience with this kind of result?
