I have oxidized one polysaccharide and make a hydrogel with Schiff base reaction with gelatin, but the hydrogel makes my medium acidic. I cannot realize what step of my work is wrong.
Hi Mohammad Sadegh Aghajanzadeh . Some gelatins are made by acid hydrolysis of collagens. You can try dissolving the gelatin in buffers that do not interfere with Schiff base formation, PBS, HEPES, etc.
Schiff's base reaction is reversible reaction and you need to go for one more step for reduction the imine bond to single bond. reducing agent such like sodium borohydride
