I wanted to use commercial Giemsa staining solution and diluted it 1:10 in pH 6.8 Weise buffer. It says to filter the solution, however, most of the stain seems to preticipate in the filter. The final working solution looks fairly dilute. The last time i used the solution i forgot to filter. The result was nice but also excessive precipitate on the slide.
Is this normal? I'm using Giemsa stock solution from Carl Roth.
You should take a look at which filter you used. What is the pore size?
Hi, I'm using a MN 615 filter, medium fast, medium retention, retention size 4-12 µm. 15 min after mixing. Before dilution, the solution passes the filter perfectly. But if I dilute it, either with A dest or buffer, either 1:10 (worse, there the filtered solution is just medium blue and transpaent) or 1:20, a lot of precipitate forms that falls out of solution. I mean, even if the filter would not retain the little crystals, how would they contribute to staining? If I use the solution unfiltered, I just get a lot of dirt on the slide. I see no difference in cell staining between May-Grünwald alone and combined with diluted Giemsa, even if I don't filter.
