No, the problem here is that signals produced by the cells themselves accumulate in the medium, and induce differentiation. The frequent media changes are more about getting rid of these, than they are about feeding the cells.

You can try less frequent changes, side by side with frequent changes, as a learning experience. They'll differentiate rapidly. It's easy to see for yourself.

Hello John Schloendorn thank you for your answer. As you said, iPSCs differentiate easily without frequent medium changes. The thing here is that, these cells are already differentiating. I’m talking about a differentiation protocol where iPSCs are forming ocular cells. All that these cells need now is to maturate. I wonder if not changing the medium for a few days would cause any problems. Looks like I’ll need to try and see.

I used to change medium every 48 hours when I was differentiating my MSCs into cartilage. The reason is simple the main growth factor in the differentiating medium will deplete after that time and need fresh. Half life of growth factors in medium, check that.
