I have some questions related to cell plating in 6-well plate.
I have plated my cells (miaPaca-2 and PANc-1 cell lines) in 6-well plates. The Media I use is DMEM (with 10% FBS and 1% Penicillin&Streptomycin). After cell plating, I gently shake cell plates to make sure that the cells are evenly spread in each well and incubate cells in 37 degree celcius, 5% CO2 Incubator.
Unfortunately, on the next day, I notice that my cells are clustering and form circular rings at the middle of the plate.
Apart from these two cell lines, my other lab mates also sometimes face the same problem with other cell lines. However, miaPaca-2 cells seriously aggreate to one another, so that I cannot use it for further experiment.
Is there anyone who has ever experienced the same problem? Or any recommendation to above issue?
It sounds like it might be a vibration issue in the incubator since your other lab mates are experiencing the same thing. Are your incubators on vibration tables?
This is a detachment problem. And your cells are known for it. As Kealan said, check for vibrations in the incubator. Also try flasks designed for easily detachable cell lines. Can you post a photo of the cells in comparison to some other cells kept in the same incubator?
This might be raelated with how you shake them. ıf you move your 6 well plate in circular directions that will make your cells come together in the center. Try to move your plate in horizontal and vertical directions after you seed your cells.
