Why do I get less DNA yield (by 2-3 times) when extracting from Jurkat cells in comparison to HeLa cells? I extract with standard Genomic DNA extraction kit.
I figured so - but it could just be this standard error you might forget when using different protocols.
I have previously observed that for other kits, the expected yield of DNA in µg differ between different cell types. One example may be found here: http://www.sigmaaldrich.com/life-science/molecular-biology/dna-and-rna-purification/mammalian-genomic-dna-miniprep-kit.html Oddly they also report that the expected amount of DNA from HeLa is about 2-3 times higher than for Jurkat cells (for the same input amount - 2x10^6). Sadly, I'm not aware of why this is so. But their reported results matches your observations quite nicely.
