We are using Mammalian Genomic DNA Miniprep Kit from Sigma Aldrich. The typical yield from 2 x 10^6 cells is around 7-8 µg, although from HeLa we get 2-3 times more. Does any one have an explanation why is it so, although these are both human cells?
And could you suggest any other extraction kit and how much DNA you can get from Jurkat?

This is most likely due to the different DNA content per cell; these are both tumor cell lines.
