I prepared Kings B agar on Friday so I could use it on Tuesday. However, when I opened the chiller, some of the agar plates had melted. There was water in the sleeves I used!
This has never happened to me and I'm really not sure what went wrong.
Has anyone had any experience with this?
Hi: If the medium frozen down it will break the agare on the medium and it will not have the ability to hardness the medium.
It could also be that the plates were warm when they went into the chiller causing condensation in the bag and plates. This could cause the agar to get wet and dilute it out the agar if it had not solidified well when first put in the chiller. I have had this happen before and only put plates in the refrigerator after they have completely cooled most times waiting over night to bag them and chill them down.
If the plates were warm & immediately kept in chiller, condensation of water will take place & moisture will be found in plate. If this moisture high & found all over the plate or media, once the grip of agar loosens it will not be attached to plate surface plus water holding capacity of agar will also matter. Agar will loosen the water it was holding.
as suggested by Tasha, put plates in the refrigerator after they have completely cooled most times waiting over night to bag them and chill them down.
