I am currently doing ex vivo work using ectocervix tissue. My tissue has a lot of cells migrating out but none of the cells are attaching on the plate. The cells seem to just be sliding on the surface. Has anyone done the explant method successfully using human ectocervix tissue? Should I coat the plates or use different media? Currently using DMEM (10% FBS) and also KSFM serum free media. Both media shows no attachment of cells.
Use FBS at 5%. In My opinión 10% could be toxic for the culture....for this reason the cells are dying.

You ma may need to provide a better surface for attachment. Are you growing them on glass or plastic? Either coat the surface with fibronectin, collagen or polylysine or alternatively you can buy culture dish inserts coated with matrix ccomponents. I don't think 10% serum is a problem, but the dish substrate as it appears, is not allowing them to adhere.
