Hello,

I am trying to isolate individual colonies using Fisher paper cloning discs. I soak the discs in Trypsin, place on top of colony, and the cells easily attach to the paper. When I transfer the disc to a new dish, I gently shake to detach the cells, but the cells will not detach. I can see cells around the edge of the disc, but none end up adhering on the bottom of the dish. I have tried trypsinizing (0.25% with EDTA) with forceful pipetting. Even tried crude mechanical removal by using forceps to scrape cells off but no luck. Anyone have an idea of what I am doing wrong?

Also, if you have any unconventional ways to isolate colonies, please share. It is driving me batty! =)
