I performed an ELISA on human serum at 8 dilutions and when interpreting the results I got drastic differences in the interpolated data between softwares. I consulted a few people who are more proficient in bio-statistics than I and have not received a satisfactory answer.
That is weird. I'm assuming that you are using the same curve fit, and it's not somehow switching between a 4-parameter versus a log/log linear, or something else? I've never used the excel plug-in. I've used just the regular curve fits in excel.
One thing I do know is that graphpad does set a min max in their interpolating. Granted, this only applies if your values are off the curve, and that is never recommended to be used anyway.
I had a similar issue with Graphpad, I just couldn't get proper data (according to the kit supplier) from the curve, so I went with the excel add-in. In my case, I got it from myassays.com. I think ELISA Excel add-ins are trustworthy as they have been developed exclusively for that purpose.
Really? I have never had an issue with graph pad. And I always do a back calculation of the standard curve to see how close it matches as a control for the curve fit.
I just guess I don't understand how the add-ins could be so different. They all using the same math. The only thing I can think of is that if you do the curve fit yourself, the concentration has to be in log10 (which means you have to do the 10^X calculation from the interpolated values). So there is an additional step on the user part. The add-ins probably takes care of that.
You are right, ELISA add-ins are more "friendly" to use and they do make the calculations automatically based on captured OD values.
