After trypsnisation cells morphology is changed within 24 hours,media was changed after cells are attached but still cells are looking unhealthy why?
You could try to use Accutase to detach you cells, I have used it with sensible cell lines since it is less damaging compared to Trypsin

What was concentration of trypsin used during the passage and how much time incubated with trypsin also effect the morphology of cells
Our Lab routinely passages A549 cells and we use Tryple to dissociate. It is much gentler on cells than Trypsin. Are you incubating with Trypsin for only 5 minutes and then neutralizing with an equal volume of medium? The only other think I can think of is maybe an issue with serum concentration? We use 10% FBS in high glucose DMEM supplemented with 1% Pen/Strep. These cells are pretty hearty, so they should grow well.
Thank you so much. I have similar A549 morphology and they grow abnormally slow. I treated them with trypsin 1c.c. for 3~5 minutes in 37'C incubator to dissociate. I will try to shorten the time and pipette them more gently.
