Our Lab routinely passages A549 cells and we use Tryple to dissociate. It is much gentler on cells than Trypsin. Are you incubating with Trypsin for only 5 minutes and then neutralizing with an equal volume of medium? The only other think I can think of is maybe an issue with serum concentration? We use 10% FBS in high glucose DMEM supplemented with 1% Pen/Strep. These cells are pretty hearty, so they should grow well.
Thank you so much. I have similar A549 morphology and they grow abnormally slow. I treated them with trypsin 1c.c. for 3~5 minutes in 37'C incubator to dissociate. I will try to shorten the time and pipette them more gently.
