In my experiment, I got a weird result, the fluorescence value of liver lysate is lower than the blank sample (with no MTP source). Did anyone encounter this kind of problem?

There are three reasons, your flourscent spectrophotometer sensitivity was set in a different spectra range, second there is a problem in ingredients or reactants of blank, third you using a different filter and lyasate color is fade thicker than the supernatant or clear plasma.

The lysate color display yellow. does it matter?In addition, I incubated this mixture in EP tube under waterbath for 3 hours, then take out and pour into white plate. It is OK? Thank you ahead.
