Hi all,
I have a problem regarding influenza virus inactivation with Formaldehyde. After incubation in cold room (4-8 ◦C), a white precipitate is formed. I haven’t got the problem before.
Have you experienced such precipitation in virus inactivation?
Which changes could have caused such a problem? Impurities or temperature changes?
Any idea about this problem?

Alternatively, you have a higher concentration of virus and/or other protein in this particular preparation and the incubation with formaldehyde caused protein aggregation when the suspension was cooled down.

Thanks for your kind answer.
The source of my formaldehyde is the same as before(before the precipitation problem ) . need to mention the same lot.
I always dilute the formaldehyde in PBS .from 35-40% to 0.2%.
my viral stock is light suspension. not completely clear. but it was always like that.
any other suggestion ?

maybe impurities . (like ovalbumin)?

It's one of the things to rule out . . . also we have seen that with 'old' 37% formaldehyde precipitates can form in the stock bottle, not sure if these would easily dissolve once they get to a lower concentration environment but I seem to remember having to heat the solution to get it to go all into solution when making it from solid stock.
