I want to make a list of 150 western blot ABs to screen for interesting mechanisms/pathways in CRC cell lines (HT29 and HCT116 which we will treat with some drugs). Which pathways should be considered? Which ABs should I look at for assessing the pathways?

There are various pathways known to associate with CRC and I would recommend you to look for following list of protein -
1. AMPK (involving Ras/Raf)
2. cMyc and cMyc target genes
3. p53 and its target genes like, MDM2, p21, Bax, PUMA followed by PARP Cleavage
4. TGFB and Smad4
5. Galectin-1,3,7 and 9
6. Nrf2 and its downstream genes in response to drug
7. WNT and B-Catenin
8. MAGE-A and p53
9. HIF-1 and 2 followed by its target genes
10. Bcl2 and related apoptotic and pro apoptotic proteins
These set of pathways would definitely help you to get understanding of CRC.
