I an undertaking tube formation matrigel assay using HUVEC cells. I've tried using 24 well plate and the images looked great! However, due to the expense of using matrigel I tried with a 48 well plate and the images didn't look that great as well as a lot of inconsistencies.
What do you guys suggest 24 or 48 well plate for matrigel assay?
Often you need to increase the amount of matrigel per cm2 the smaller the well size. Therefore, if you use 50 ul matrigel per cm2 for 24 well, consider increasing to 100 ul matrigel per cm2 for 48-well/96-well.
However, my recommendation is to always use the best conditions with the least amount of inconsistencies as this data will be much better - also, pretty images!

There is nothing as such. You can use 24- or 48- or 96-well format. All you need to do is optimize the protocol.
The tube formation matrigel assay needs a well-defined thickness of the gel layer. The basement membrane surface and its components play an important role in tube formation.
Make sure that the cells are healthy, and you also need to optimize the seeding density.
The disadvantage of this assay is that there is large variation of tube-forming capability among different lots of endothelial cells and support matrices.
