I didn't see the use of a mix of cell wall degradation enzymes in the description of your method.  It is not really a squash method once the cell walls are gone, which makes the chromosomes to be spread easily. You can check out any one of the Arabidopsis papers on chromosome spread for meiotic studies for details of the method.

 I have not tried the squash technique,  Do you heat the slide before squash?   You may ask chromosome researchers.

Hi, I generally squash the anthers fixed in carnoy for one night at +4. You may also store them in 70% alcohol for several months after fixation. If the anther is small you dont need to hydrolization step (1 N HCl -60 C- 5 min). I directly squash the anther in 2% orcein solution gently. It is also important to take the anthers in the correct time of the day. 

Try warmed slightly (60 degrees) in an incubator, then it is easier to separates. Orcein is a good dye for species that have little chromatin. Also this dye can try. Good luck

After staining of fresh, crushed anther with  2% aceto orcein (60degrees) we get the best results

Try using very young anthers or pollen cones (this depends on species used...I used maize and am now trying out on cycads (Zamia species)...with flowering plants, put directly in Farmer or Carnoy and with cycads try pretreatment at 1°C for 24 hours, then put in fixative...try collecting at 11:30 a. m. or  1:30 p.m...furthermore, just type the question on the internet and lots of papers dealing with the subject will be available to you...remember methods may vary depending on species, time of collection and age of simple...luck
