Dear Experts! The culture vessel and ventilation types used in tissue culture set the limits between the plants grown in the culture vessels and their external environment. Nevertheless, the type of vessel sealing may determine the efficiency of the propagation technique. I would like to know that which type of ventilation is suitable for the propagation of woody plants in vitro?

Dear M. Manokari it depends mainly from species. I've worked with 2 different tree species: walnut and cherry. These're completely different in their in vitro behavior. Whereas cherry might support several closure conditions, walnuts are less plastic. Walnuts usually don't support tightly closed vessels, having a suitable behavior with vessels (caps) that favor gas exchange. Even in TISs, at least under my conditions, was necessary the introduction of extra aerations to obtain high quality microshoots. you should also check the surface temperature of culture media. overheating due to the light source is also another impact and can be integrated with ventilation and may result in Vitrification depending on explant source and cytokinin concentration in medium formula.
Like to use suncaps on tubes and magenta boxes for hardening phase of rooted shoots.

From my experience, forced ventilation system might help woody plants grow better. and also help us save time and labor cost in large-scale micropropagation. We used this system for Paulownia fortunei and Thyrsostachys siamensis. The ventilation rate will depend on plant species and can be controlled by using air flow rate meter.
Sorry! I want to add one more thing to my previous answer. The forced ventilation system should go with large culture vessels containing hundreds of cultured explants.
