I am using Tissue Lyser with Ripa Lyzis Buffer to obtain protein samples from certain tissues for western blotting. I wanna enhance the recovery of total protein from the samples. Which type and size of beads should i use for homogenizing the large intestine tissue of rat?
You can use zirconium beads which are commercially available, or alternatively simply grind broken glass and perform sieving to get more or less similar sized glass particles which may be acid washed overnight in 6N HCl, followed by washing with distilled water and thence they can be used to homogenize tissue in a homogenizer.

I honestly just use a 2.4mm steel bead for most tissue types, but I just varying the intensity and the time based on whether it is soft tissue or hard tissue.
