General PDA media is best for the isolation of soil fungi. I have been working four months on isolation of fungi from soil. I got the best results and recommend the PDA media.
Sabouraud's Dextrose Agar is usually used for the isolation of clinical fungi. So, Potatoes Dextrose Agar is more suitable for the isolation of environmental and saprophytic fungi. I advised you to use PDA prepared with inclusion of cycloheximide and chloramphenicol in order to inhibit overgrowth of unwanted fungi and contaminating bacteria.
I supose you are interested on filamentous fungi. In that case PDA better than SAB however, depending if you are screening samples looking for some specific genera, you should try other media, such water agar (WA) or DRBC (Dicloran Rose Bengale), the last avoid invasive growth. In order to no loose biodiversity (in case you are looking for) you should try different media in parallel.
PDA is a general media for the enumeration whereas sabouraud has certain specificity.
While doing isolation the purpose is to get more & more different types of fungus hence PDA is ideal whereas comparison is always better at least you will get a basic data in hand.
you may add soil extract in PDA to improve the number of isolate and make the chemical environment more conducive for the the culture dependent soil isolate.
You can use PDA or SDA with chloramphenicol to recover common environmental fungi like Aspergillus, Fusarium, Chaetomium, Mucor, Rhizomucor, Alternaria, Rhizopus etc.If you wish to isolate dermatophytes such as Microsporum gypseum, you should use SDA with chloramphenicol and cycloheximide. However, for isolation of Cryptococcus neoformans, Candida, Geotrichum, Rhototorula from soil, avian droppings, bat guano, air, fruits and vegetables, we employed Pal sunflower seed medium with chloramphenicol.This medium was developed by us in 1980 and is used in many laboratories.We developed one more medium designated as "APRM" in 2015 and is helpful to recover common saprophytic fungi from soil, avian excreta,water, and air.I have uploaded several papers on Research Gate and Academia on the efficacy of Pal sunflower seed medium to isolate many fungi from environmental and clinical specimens.
