For what purpose are your trying to grow the jurkat cells, Heshu? To test against a certain genetic mutation or disease? I do not have the reference but I understand Biotin helps them proliferate and there are certain other enzymes that have a proliferative effect.
Excellent, Heshu--the Biotin might do what you need. I had not thought of making them live longer because they are so robust and reproduce so well already. Something to think about is associating Kreb's Cycle activity relative to anticancer activity and Jurkat cells. That would involve, if you are equipped (I am not) to measure oxygen and pH levels of the cells. We see it anecdotally with dietary changes in cancer patients, but have not conducted an experiment specifically on it. Cell entropy experiments have touched upon the concept, by the way. Normally, it will be better to ask the information from the company who sold the cells to you or the previous cell owner. And I cultured my Jurkat cells in RPMI1640-10% FBS.
My Jurkat cells were maintained in RPMI 1640 medium supplemented with 10% heat-inactivated fetal calf serum (FCS), 1mM L-glutamine, and 50g/ml penicillin/streptomycin. The culture medium was renewed every three days bringing the cell concentration to ~2x105 cells/ml.
after adding plasmocin during two weeks, these cells proliferate in RPMI 1640 10% heat-inactivated SFB 100 U/uL Peni/Strepto and 2 mM Glutamine. Life Technologies provide an useful RPMI (GIBCO) with HEPES.
I have a problem relating to Jukart cell line myseof. I hope someone helps me out. Whenever I make duplicates from same of Jukart cell line and count them the next day, I often do not get the same or even cell density for the cells. Any advice?
