I am trying to isolate cellulose degrading bacteria but it isolates are not able to degrade cellulose potentially. So I need a suggestion which source is best and which method is best for isolation as well as which media is prefer?
termites are the best source of them (rumen content contains them too, but as much - and then better to take the half-solid fraction, not the very fluid)
in general, you can focus on bacteria from intestine of animals that eat more woody plants; or from environment where plants are degraded
Decaying litter, leaves, cotton plant, newspapers are the best natural habitats where you can find aerobic cellulose degraders. Use 2% carboxy methyl cellulose + 1% glucose to isolate cellulolytic fungi and 2% CMC + 0.5% beef extract for isolating cellulolytic baceria. Use 10000 time dilution of samples as disscussed above preferably by spread plate method (details can be found in Practical Microbiology by Cappuchino and Sherman),
