I tried to dissolve the foot lipid extract from a mollusk in DMSO, however it seems to be insoluble in DMSO even in low concentrations. Could you please suggest any other solvent which could dissolve lipid extracts and must be safe for cell culture and doesn't interfere with the anti-inflammatory results.

Try methanol to dissolve the lipid extracts. In our lab, we are using methanol for lipid extracted from fish for GC-MS analysis.

methanol is not recommended for cell culture as it affects the viability of the cells and gives false results.

Depends a bit on the cells you use..It may be advisable to test your cells beforehand with a parameter like inflammation markers, viability staining, LDH release, apoptosis or similar, with different solvents and different concentrations. For shorter studies we routinely use up to 1 % ethanol on "tough" cell lines like Caco-2. But you may also have a solubility issue as such, i.e. can you solubilize your compound at hand in the correct concentration. You should probably have some srum present, in order to "buffer" your lipid in solution, along with a solvent like ethanol. But start with control experiments :)

i use RAW264.7 macrophages and 3T3 swiss albino fibroblast cell lines. i test RAW for NO and TNF inhibition and 3T3 for PGE2 inhibition assays.
