We have recently extracted the grape seeds with solvent mixture composed of acetone:water:acetic acid(90:9.5:0.5). Which solvent will be used or appropriate to make different dilutions of aforementioned crud extract for antioxidant assays.
Hello, best solvent for antioxidant extraction from grape seed is ethanol/water mixture with ratio of ethanol to water 1:1 to 3:2 (v/v). A higher extraction yield will be at temperature probably 45*C and ratio of liquid to solid 7-8: 1 and extraction time around 3 h.
But according to jay parkash et al they have extracted the grape seed in acetone water acetic acid mixture, but they haven,t mention with which solvent was used for further dilution of their extract used in analysis.
For an unbiased determination of antioxidant capacity, I will recommend that you first evaporate the solvents used for extraction and possibly dry the crude extract. You can reconstitute in one solvent in this case methanol or ethanol. This is very important especially for some antioxidant capacity assays like DPPH and FRAP!!!
