After you check out the new codons with a software, are you going to order the new designed optimized cotton gene sequence from a company for expression experiment in corn?
If so, why don't you just check out a company for service? The service will include software searching. And, I think the price shouldn't have that much different between 'you submit your newly designed gene for synthesis' and 'submit the old sequence for them to do the whole thing of codon-optimization'.
Thanks Yaun-yeu yau, but i have optimized it as i want. i am satisfied that is why. The purpose was not much high expression and i created codon usage and codon context myself with selected gene.
Yes, I understand, if you just want to learn and practice. My question is: after you have the new codon-optimized gene sequence on a sheet of paper, what are you going to do with it? Are you going to synthesize it yourself in the lab or are you sending it out to a company for synthesis, so you will have the 'materials' for cloning?
