You could try with the configuration that is most likely to work with tumors. It will also depend on the invasion potential of the tumor cells you are working with, but a good starting point is a wound healing assay with 0.1 mg/mL high molecular weight HA ( MW: 1.46 MDa, Viscosity: 2.3 M3/kg). You can measure the wound width at 0, 8, 24, 32, and 48 hr. You will most likely bring the FBS down to 1% if you cells tolerate it.

Thanks Leonardo. Is there any provider that you would suggest to order HA? Additionally, I have some more questions about the HA wound healing assay. I guess this high molecular weight HA will behave in a viscous and gelified form, won't it? In this case, I think it'd be used as a coating in the culture wells, and then adding the cells. I'm using silicone inserts to create the wound, so even if I coat the whole well, then attach the insert and then seed the cells inside, I was wondering if the HA coating would be removed/damaged when I lift the insert. On the other hand, if I'd go for a more rough wound healing assay, using a tip to create the wound, the problem with damaging the coating is even more likely to occur, isn't it?
