Tail protocol or tissue protocol? Any recommendation for this procedure? I need to extract DNA from an ear punch of rodents with a commercial kit from promega.
What downstream processing you need to do with the DNA? If you want a crude prep which can be used for PCR, genotyping etc use the hotshot method. Its easy and quick.
I use the Phire Animal Tissue Direct PCR Kit from Therom Scientific.
I incubate the ear punches in 20ul TE buffer (pH 8.0) with 0.5ul DNA release additive. This is vortexed for 15 seconds whereafter I incubate the punches for 5 minutes at 75 degrees en for 2 minutes at 96 degrees.
After the incubation I put the punches for 10 minutes in a centrifuge (13000rpm) and pipet the supernatant in a new eppendorf (0.5 ml). This is the sample DNA I use for PCR. I works well for my genotyping.
