Hello,

does anyone have any experiences with transferring the SV40 origin as independent functional element to another vector/plasmid?

I would like to transfer SV40-ori from the pEGFP-N2 plasmid to another vector to ensure its episomal maintenance in human cells but after studying the literature I have noticed that SV40ori-adjacent SV40 promoter and enhancer elements significantly influence replication efficiency (Bergsma et al.,  Proc Natl Acad Sci U S A 1982: http://www.ncbi.nlm.nih.gov/pmc/articles/PMC345744/ and Chandrasekharappa & Subramanian, J Virol 1987: http://www.ncbi.nlm.nih.gov/pmc/articles/PMC255869/). Now I am wondering if it is vital to transfer all the adjacent promoter/enhancer elements (72 bp- and 21 bp-repeats) together with SV40ori to achieve optimal replication of target vector or would it be sufficient to transfer only the region comprising all three 21 bp-repeats together with SV40 core origin?

Thank you,

Peter
