Dear Researcher,
I am investigating new therapeutic approaches to treat Glioblastoma. By testing my approach, I found that in one mouse model the data are promising. However, I am looking for validating my data. I would like to re-do the experiment using a different mouse model for the same disease.
I know that the best is to test the treatment approaches on two different mouse models (two independent experiments each) However, due to running out from the funding and time I must choose between two options.
Either using one mouse model (two independent experiments) or two mouse models (one independent experiment each).

Hi, It is always good to show the drug effect in multiple mouse models. Also if your mouse model represents the most aggressive form of the disease then you can focus on that. Thanks.
I think it would also depend on the variability of the results so far. You may need to increase the number of animals, dosage range etc.
if you have statistically significant results already in one model then extension to another model would be valuable.

I would definitely go for one experiment in two different mouse models. Usually if you use enough animals you don't need another independent trial to confirm your results (i.e. if they were significant). Showing efficacy in two different models, or differential activities in two GBM subtypes will have a much greater impact than using one model.

The mouse models are most used due to several factors such as high sensitivity to the treatment, easy to manipulate and also require less administration volume of active principles.
Yes you can increase in number of animals (Male and female) from 2 species as for example Bal C, but most of the time females are most sensitive. In addition, you can also used a range of dose.
You need to do a Power Analysis to ensure that a negative result is real and not just a result of too few animals. If sufficiently powered, a single experiment in each of two strains would be more meaningful than 2 experiments in 1 strain.
I think it's better to have solid data on one mouse model (meaning two independent experiments work consistently), and if it works you can use this data to propose for funding using the second model. Of course, you have to consider seriously which one to start depending on priority.
