We will use human microglial HMC3 cell line in our experiments. It's written in ATCC that EMEM medium must be used for culturing but DMEM/F-12 medium was used in some articles. So, which medium is better for that? Could we use DMEM or DMEM-F12 instead of EMEM?

When I used these cells, we used the Eagle’s minimum essential medium (EMEM), containing 6 g/l glucose, 10% fetal calf serum (FCS), 2 mM glutamine, and antibiotics.
It's not an easy cell to work with.

we had difficulties because the cells had a little slow growth, and were not responding well to LPS. But after some time in cultivation, they began to respond well. We found that they are very sensitive to temperature changes.

I have been using the HMC3 since couple of months now, they have a PDT of 32-35 hours with EMEM (Eagle's MEM) + 10% heat inactivated FBS + 1% P/S + NEAA + Sodium Pyruvate.
After several attempts, we managed to detect IL-6 induction after costimulation with LPS and IFN-y, s it was already described by Cinzia dello Russo in an other discussion field:
