I have been using a Pierce BCA Protein Assay kit (Thermo Scientific) and diluting my samples (saliva:diluent) 1:10, 1:20, 1:40. I do a triplicate of each dilution. The variation in between triplicates is negligible; however, when calculating the concentrations for the same sample, I get significantly different values for each dilution. Also, I decided to test an undiluted sample and the concentration I obtained was (while inside the range of my calibration curve) nowhere near the 1:10 dilution.
Do you know of a better assay for saliva supernatant? Additionally, should I consider not diluting my samples that much?
