Dear friends and colleagues .... I thought to ask here in case any one has a previous experience with any of the following electroporator systems, Which is better for mammalian cell transfection?????? we are supposed to eb using it for stem cells, as well as various mammalian cell lines:
1- ThermoScientific Neon System
2- BioRad electroporation System
If anyone has a previous experience with any of these systems, would really appreciate if he/she shared it with me. Any piece of info/feedback (positive or negative) based on practical experience is really appreciated!

I've had a little experience with the neon system, but have not used the biorad system. Overall the neon system is effective, but only if you have the time to troubleshoot. The system comes with information on what works well for some cell types, but often these conditions are not always the best. Sometimes you will have to troubleshoot and fiddle with the conditions to get it just right, and it can take a decent amount of time. There are other times that will just take a few days to get it working right.
It also depends on what you are transfecting. Plasmids will be difficult and can require additional various methods of tweaking, whether it's electroporation conditions or adjusting cell count or plasmid amound. Small molecules like siRNA are very easy to transfect.
I have had experience in using to transfect HEK293, immortalized human brain endothelial cells, sh sy5y, and primary mouse brain endothelial cells. I have not done stem cells.
One thing about the neon system is that the expensive buffers are easy to remake and the expensive tips and tubes can be clean/sterilized and reused, despite thermo saying otherwise. There are other answers on researchgate regarding these things if you were to go with the neon system. Let me know if you do purchase and are having trouble with cell viability/transfection efficiency!

As an inventor of the NEON electroporation system, I would say it works very well with most cell types. All electroporation systems that uses cuvette, kill cells massively due to the large electrode surface area.
