I want to test IL10 secretion from B lymphocyte. Do I need to use Monensin or Brefeldin A for blocking? Kindly clarify whether or not Monensin is a B lymphocyte blocker and Brefeldin A is for T lymphocyte blocker? Or can any of them be used for blocking B cell?
Both of them block secretion of cytokines from cells (interacting with golgi and inhibiting protein transport-momensin) or redirecting the proteins to ER ( brefeldin A). If you want to to see IL10 in human either would be ok i think.
The publication below clarifies some of their properties. Brefeldin-A is generally used to halt secretion of proteins by disrupting Golgi to transGolgi transport (http://jcb.rupress.org/content/116/5/1071.full.pdf). Monensin and Brefeldin-A are both used in intracellular cytokine measurements.
I do not think so. These regents are very toxic and should be used at doses recommended for cells in vitro.
