Which gradient and ultracentrifugation settings are the most relevant to purify CVB3 and SARS-CoV 2?

Hello,

I am looking to purify Coxsackievirus B3 (CVB3) and SARS-CoV 2 viruses by ultracentrifugation (Optima XPN from Beckman Coulter) and after several searches in the literature, I decided to use an iodixanol density gradient (OptiPrep). However, I find very different gradient protocols and ultracentrifugation settings and I am therefore lost. Could you tell me which gradient and ultracentrifugation settings would be the most adequate to purify CVB3 and SARS-CoV 2?

First are both viruses together? If not do some experiments with each separately you know like in science research you learned in school. If they are together do step1 for each as before then try to separate the mixture. The attachment may. be of some assistance. Best wishes, an old Model E jock.
