I'm working on GUS detection whereby I use MUG as substrate that will be cleaved by plant extracted protein to produce fluorescence compound (GUS fluorometry method).

In the protocol, i mix plant extracted protein with MUG substrate, incubate at 37C, then take 100ul mixture out and add to 900ul stop reagent (sodium carbonate 0.2M). This will be transferred to cuvette and measured at excitement 365nm and emission 455nm.

Questions:

1) As our equipment for measuring fluorescence can only use cuvette (cannot use microplate), can anyone suggest a suitable type of cuvette to use? I know that quartz can be used but its too expensive. Any other types of cuvette that can be used?

2) Can disposable PS cuvette be used? I have a lot of samples, so if I can use disposable cuvette (and maybe reuse it again after washing), then I can measure my samples faster instead of using one cuvette whereby I have to spend time to wash it between samples. Since PS disposable cuvette (macro type 4.5ml capacity, 4 sides clear) can be used for 340-780nm, can it be used for fluorescence detection? Can disposable ps semi-micro cuvette be used for fluorescence detection as well?
