I'm going to use phage display for selection of high-affinity peptides against some cancer-specific receptor targets. As I'm not using the commercially available premade phage libraries, I have to construct the library on my own. To this end, I need to clone a randomized library of insert DNAs (encoding the randomized library of peptides) into a phage vector. Now, I have to decide which of the above-mentioned phage display systems I'm going to choose! If you have any experience or knowledge about this, I'd really appreciate sharing your opinions.

Dear Farzad,
I am also working on phage display library. I want to know that have you confirmed your peptide, number of mutations and position? If so you can design your primers according to that and proceed for display with the help of Helper phage M13. It will be the best system for display and also it will be easy for further screening against the target.
