Hi everyone, I want to investigate the expression of two genes (GPR120 and PPAR-γ) in PBMCs. Now I want to know how much of the two genes are in leukocytes and which cells express this two genes?
The GPR120 gene has two isoforms (short and long). Which isoform of this gene is more expressed in leukocytes? Are both isoforms express in leukocytes?
The PPAR-γ gene has 6 isoforms (PPAR-γ1-γ6) and 10 variants (var1-10). Which isoform/isoforms of PPAR-γ is more expressed in leukocytes?

It depends from the target species. Are you talking about human ? The expression of these two genes is relatively low in human leukocytes compared with other human cell types (tissues and organs). You can find information in the expression atlas:

It is also known that the expression in human PBMC could be changed dramatically after treatment with certain compounds.

Yes, I am talking about human. My samples is venous blood, and I extracted total RNA by column method. After the cDNA synthesis by Takara kit, I twice designed specific primers for each of the genes (GPR120 and PPAR-γ), but unfortunately none of two designed primers do not respond (I cannot amplify these genes by PCR). The housekeeping gene (GAPDH), as a positive control, always responds along with my tests. What is the problem?
I agree that the expression of these genes in the leukocytes is low, but they should be amplified by PCR.
What compounds or drugs can reduce the expression of these genes? Can corticosteroids impress the expression of these genes?
Please guide me.
