It is a while that I'm trying to recapitulate the literature for IRF5 and/or IRF7 nuclear translocation and/or phosphorylation upon TLR3/ 4/ 7 stimulation in undifferentiated or PMA-differentiated THP-1 cells; but, I have not been able to detect neither of the mentioned phenomena. For the time being; I do not care about the cell line or the method of stimulation at all. I'm just looking for a cell and condition that would obviously show me either of these two changes in IRF5 and/or 7 status.
The information in the paper in the link addresses your questions. Murine cell line in response to activation of TLR7 and 8 induced signaling activated IRF5 and IRF7, but not IRF3.

I forgot to mention that I mean "human cells"; and since me and several other Labs have failed to recapitulate the published data; I am really looking for someone that has tried the mentioned analyses her/himself so that I would be able to discuss about their protocols in a very detailed manner in case that I'm missing any tiny but pivotal point. so, I would really appreciate if you put me in contact if you know such a scientist...
