maybe someone with experience in the field of cytotoxicity testing of materials for medical use may help me. I have two possible cell lines available in my lab: mrc-5 and 3T3 fibroblasts. According with the ISO 10993-5 guide I should use

experts to be suitable”
I don’t have the A31 clone of 3T3 cells, would you recommend me to use MRC-5 cells instead? Or regular 3T3 cells may be useful for this kind of evaluation?

The choice of cell type for testing the cytotoxicity of any biomaterials including nanomaterials depends on the target organ and the route of exposure.
For example, I work with chitosan nanoparticles (CHNPs). Initially, I tested the cytotoxicity of CHNPs on HEK293T cells and obtained a minimum inhibitory concentration. The same concentration didn't work when I tested it with my target tissue cell type ( Mesenchymal stem cells). It is recommended to test the biomaterial with the target organ cell type to arrive at a more significant conclusion.
If MRC-5 cell line is similar to your target organ cell type, I think you should go ahead with that, or any other avaiable cell line which is similar to your target organ.
