I want to detect effect of a bacterial toxin in human cell line and want to see cell viablity, so which assay will be better, many people critisize MTT.

You can use cck-8( cell counting kit-8) which is an alternative to MTT.

You can use also neutral red as an alternative to MTT. Then, there are more sensitive bioluminescence assays such as CellTiterGlo which measures ATP content or Real-TimeGlo viability assay, which you can use to monitor cell viability in time up to 72h.
Personally, I use MTT assay without any problems.

I'm just confused between viability and prolefiration of cell. Are both of them samef different? Do both of these phenomena require same of different assays?
Most of assays are based on the metabolic activity of cells or membrane permeability. These based on membrane permeability are used only for cell viability (dye enters only to non-viable cells). These related to the cell metabolism may be used for both purposes, because metabolic activity increases proportionally with the cell number (proliferation) and you can also monitor the toxicity of the treatment (viability).
I used MTT for both, cell viability after treatments and cell proliferation in time course experiments.
You can perform BrDU assay (very sensitive) or WST assay (more sensitive than MTT)
CCK assay/cck8/WST assay is the best assay to measure cell viability
Beside viability assay. Please check your lysis time also? during your isolation process. All existing cell viability assays are good but whatever works for your cell type stick with that. TRYPAN BLUE is the best for quick and inexpensive. Try to use 0.4% stock, final 0.2% (1:1ratio) and look under microscope under low and high power for your satisfaction. YES, all these above mentioned kits/reagents (such as CCK-8/WST/ATP/MTT) have their pros and cones. More later as needed...good luck. T
